Physiological Characterisitcs

Dextrose broth with phenol red/Durham tube         KIA agar slant (hydrogen sulfide production)
(acid/gas production)

Tube on left is positive for acid and gas production            Tube on left = uninoculated control
Tube on right = uninoculated control                                  Middle = positive result
                                                                                        Right = negative result

Starch hydrolysis on starch agar plate (tested with iodine)

Plate on left is negative                         Plate above - colony on left side of plate
(no clearing around colonies)                = positive for starch hydrolysis

Simmons citrate agar (citrate utilization)            Urea broth (urea hydrolysis)

Tube on left is positive                                        Tube on left is positive
Tube on right is negative                                     Tube on right is negative

Lactose broth with Durham tube showing gas production
(indicative of coliforms)

Levine EMB agar streaked with E. coli                Nucleated colonies on Levines EMB agar
(note metallic green sheen)

Beta hemolysis as seen on blood agar                            VP test results (left = negative, right = positive)
(note clearing around colonies)

Catalase positive reaction (note bubbles)                        Oxidase reaction (note dark blue color
of colony due to oxidase reagent)

Indole test

Tube on right is positive for indole test (Kovacs reagent)

Staphylococcus aureus on Mannitol Salt Agar

Antibiotic susceptibility testing (Kirby-Bauer Test)
